MMP-9: A Key Regulator of Tissue Regeneration and Destruction

MMP-9 is a secreted endopeptidase that degrades the extracellular matrix
(ECM). Utilizing substrates such as type IV collagen, it is physiologically involved in processes including wound healing, angiogenesis, and synaptic plasticity. Conversely, dysregulation of MMP-9 expression and activity is known to drive various pathological conditions. In oncology, it promotes invasion, metastasis, and tumor angiogenesis, whereas in cardiovascular and cerebrovascular contexts, it is associated with atherosclerotic plaque rupture and blood-brain barrier (BBB) disruption. Furthermore, MMP-9 is deeply implicated in the progression of chronic inflammatory diseases, such as rheumatoid arthritis and inflammatory bowel disease (IBD), as well as neurodegenerative and psychiatric disorders. Consequently, it not only holds the key to elucidating disease mechanisms but is also widely investigated as a potential therapeutic target and biomarker. Antibody #85604 is suitable for WB, IP, and IHC-P applications using mouse and rat samples.

Islet-1: A Key Determinant of Multi-Lineage Cell Fate

Islet-1 (Isl1) is a LIM-homeobox transcription factor that functions as a master regulator driving multi-lineage cell fate determination. Although initially identified as an insulin gene enhancer-binding protein in pancreatic beta-cells, it plays indispensable developmental roles in spinal motor neuron specification as well as the morphogenesis of the pancreas and pituitary gland. Notably, within the cardiovascular system, Islet-1 regulates the maintenance, proliferation, and differentiation of multipotent cardiac progenitor cells in the second heart field (SHF), serving as an essential factor for the structural assembly of the right ventricle and cardiac outflow tract. Mutations or aberrant expression of Islet-1 are linked to the pathogenesis of congenital heart diseases, including Tetralogy of Fallot, as well as diabetes mellitus accompanied by impaired insulin secretion. Furthermore, Islet-1 has garnered significant attention as a histopathological diagnostic biomarker
for neuroendocrine tumors (NETs) and as a primary target for myocardial and pancreatic cell reprogramming in regenerative medicine. #68324 is suitable for WB, IP, IF-F, and IF-IC utilizing human, mouse, and rat samples

Effective November 20, 2025, protocols have been updated to remove the low-speed spin step (1000 × g), streamlining the workflow and saving time. Our testing has shown no impact on performance or yield. Kits using orange-tinted Monarch Spin Columns S2C (NEB #T3017) are encouraged to follow the updated protocols for improved efficiency.

  • Extract and purify genomic DNA from a broad range of sample types, such as cells, blood, tissue, bacteria, yeast, saliva, swabs with just this single kit. Suitable for clinically-relevant samples (e.g., saliva, cheek).
  • Isolate high molecular weight gDNA (peak size typically ≥ 50 kb) for use directly in downstream applications, including PCR, qPCR/RT-qPCR, NGS and sequencing.
  • Obtain excellent yields of highly-pure DNA with very low residual RNA contamination (typically <1%) with included RNase A and Proteinase K.
  • Protocol compatible for cleanup of gDNA from other purifications.
